Philomena (Judi Dench) werd in 1952 als tiener in Ierland zwanger en werd naar het klooster van Roscrea gestuurd om te worden verzorgd als een "gevallen vrouw". Toen haar baby nog maar een peuter was, werd hij door de nonnen meegenomen voor adoptie in Amerika.
Philomena zocht de volgende vijftig jaar naar hem, maar zonder succes. Toen ontmoette ze Martin Sixsmith (Steve Coogan), een vermoeide politieke journalist die toevallig geïntrigeerd was door haar verhaal. Samen vertrokken ze naar Amerika op een reis die niet alleen het buitengewone verhaal van Philomena's zoon zou onthullen, maar ook een onverwacht hechte band zou scheppen tussen Philomena en Martin.
De film is een meeslepend verhaal over menselijke liefde en verlies dat uiteindelijk het leven viert en laat zien dat er zelfs op de donkerste plaatsen wordt gelachen.
