De film volgt Sophie (Ruby Barnhill) terwijl ze door de GVR uit haar weeshuis wordt weggevoerd en naar Giant Country wordt gebracht, waar ze onmiddellijk het risico loopt te worden opgegeten door de andere, grotere reuzen, waaronder Fleshlumpeater (Jemaine Clement) en Bloodbottler (Bill Hader).
Samen gaan Sophie en de GVR op avontuur naar Dream Country, waar ze dromen vastleggen die de GVR aan alle slapende kinderen van de mensenwereld kan geven.
Nadat ze een sluw plan hebben bedacht, gaan ze terug naar Londen om te zien of de koningin (Penelope Wilton) hen kan helpen het probleem van de pestende, kinderetende reuzen aan te pakken.
