1846 New York et les tensions ont atteint des proportions culminantes et fatales entre les immigrés irlandais et les habitants britanniques et hollandais du quartier de Five Points. William Cutting (Day-Lewis), alias Bill the Butcher, dirige les habitants de Five Points et crée le gang des « Native Americans », tandis que les immigrants sont dirigés par le prêtre Vallon (Liam Neeson) et forment « The Dead Rabbits ».
Après un violent affrontement entre les deux gangs qui laisse Vallon mort, son fils Amsterdam disparaît pour réapparaître en 1862 sous un nouveau nom et se venger de la mort de son père. Amsterdam (DiCaprio) parvient à s'intégrer dans le cercle intime corrompu de Bill le Boucher, qui dirige désormais toute la pègre autour de Five Points, et devient même son bras droit. Cependant, son identité est bientôt révélée et une confrontation sanglante s'ensuit.
